You have extracted protease from culture filtrate of Termitomyces clypeatusgrown in potato dextrose broth.During partial characterization, youchecked protease activity at different temperature, pH. Surprisingly, the activity of proteolytic enzymein some isolates increased in presence of 0.6 mM PMSF inhibitor (serineprotease inhibitor). Later, you increased concentration to 6 mM, did not see reduction in activity. The same was observed in 0.1% gelatin zymography(activity staining) as well. However, inhibition of activity was observedwith metalloprotease and aspartic protease inhibitors EDTA and Pepstatin A respectively. Catalogue possible explanation of increase in activity in presence of PMSF

The enzyme might be degraded by serine proteases present in the extract. However, PMSF inhibits these proteases increasing the extracted protease activity.
