Autoclave is used to sterilise solutions using high temperature and pressure.

As PBS is used in various assays (immunocytochemical and immunofluorescent staining, cell culture, protein purification, etc.), it is important to avoid any bacterial and fungal contamination of the solution as these organisms can impair the properties of the buffer while their presence can significantly affect the results of the analysis. As a result, autoclave is required to sterilise the PBS solution.
