Using light microscopy you examine the soil samples and the goo from the degraded polyurethane. Will this approach allow you to observe all microorganisms present in the sample ? why or why not ? What are the limitations of this approach?

All microorganisms can't be observed because limitation fortheir size likely to be present. Smaller microorganisms will not be observed with high resolution. In addition, the sample is rather large and light cannot penetrate through it. Thus, some microorganisms will not be observed at all. It is a limitation of the light microscopy that thin layers of material can be examined only. In addition, they should be transparent.
